Separation and purification of large biomolecules

The Tantti® Monolith hyperporous polymer is specifically designed for downstream separation and purification of large biomolecules – particularly in the production of biologics (e.g. protein drugs, vaccines, antibodies etc.).

Tantti® Monoliths are now available as DuloCore™ pre-packed columns. The key differentiating characteristic, in comparison to traditional purification columns, is its highly connected macropore structure that confers superior porosity, which is ideal for purifying large biomolecules at high loads:

  • Minimise the shear stress from Eddy currents affecting yield of biomolecules
  • Enables higher flow rates
  • Lower back pressure

Tantti DuloCore IEX Column

Tantti® DuloCore™ represents a breakthrough in liquid chromatographic technology. It can be used for the separation and purification of large biomolecules, with applications in production of biopharma products for advanced therapies.

IEX Chroma

Tantti® DuloCore™ ion exchange chromatography column is a next-generation polymer-based packed column tailored for the purification of biomolecules, such as DNA, virus, exosomes, proteins, and peptides. Its unique monolithic resin with macro interconnected channels enables high throughput and offers superior bioseparation efficiency, especially in the manufacture of vaccines and viral vectors.

Tantti® provides its Dulocore™ IEX columns in two different chemistries and in two column sizes, which allows users to rapidly screen and optimise purification parameters. With DuloCore™’s customizable pore size and excellent hydraulic permeability, direct mass transfer through convective flow is readily achieved.

Key applications

The production of viral vectors or viral particles has become increasingly important in vaccine development, gene therapy, and cancer therapy.

Viral particles and genetic chromosomes are large and complex structures. Furthermore, the surface variability of viruses adds to the difficulty in production and purification steps. Traditional purification, which requires ultracentrifugation, precipitation, and filtration, has disadvantages such as low yield and inconsistent quality and quantity. To overcome these challenges, liquid chromatography has become the preferred technology for viral particle recovery and impurity removal in vaccine and viral vector production.

Tantti® bioscaffolds for 3D cell culture

Applications of 3D bio-scaffold in biomedical science

The 3D bio-scaffold plays an important role in tissue engineering, oncology, regenerative medicine, drug screening and stem cell biology study.

  • To engineer an artificial organ, cells express similar behaviors, such as cell proliferation, ECM production, cell-cell interaction and cell-ECM interaction with those are in vivo through a 3D bio-scaffold.
  • For drug screening, to avoid the efficacy-prediction error and side effects, an appropriate model is necessary for drug evaluation.
  • Furthermore, the 3D scaffolds as an in vitro model are also used for drug evaluation and long-term clinical effects. 

UniTantrix™Microcarrier

What is UniTantrix™ Microcarrier?

  • The UniTantrix microcarrier provides a 3D construct for anchorage-dependent cells attachment and proliferation. It is recommended that 3D microcarrier culture is supporting the option of replacing 2D monolayer culture for cell expansion.
  • Microcarriers are made of either natural materials, such as gelatin, collagen and cellulose or synthetic materials, such as polystyrene(PS), polyhydroxyethylmethacrylate(pHEMA) and DEAE-dextran. Natural materials or its derivates with better biocompatibility, softer mechanical properties and higher reproducibility variation than synthetic materials.
  • In addition, lacking binding sites or functional groups of synthetic materials for cell adhesion is one of the major issues for cell expansion. Therefore, natural materials-based microcarrier has recently become more popular.

Features of UniTrantrix™

  • UniTantrix™ microcarrier is a unique macroporous construct with highly connectivity uniform pores and numerous surface area (6000 cm2/g).
  • It is manufactured by medical-grade gelatin providing flexible mechanical properties and stability.
  • The highly connective and uniform pores allow cells to adhere, proliferate, migrate and differentiate and the nutrients, oxygens and metabolic waste exchange effectively from the external to the internal microcarrier.
  • The macroporous microcarrier as a house-like structure to cells for protecting cells from shear stress exposure.

In addition, the microcarrier is dissolved easily by specific enzymes and the cell recovery rate is almost 100%.

UniTantrix™ Microcarrier Applications in Bio-Medical Science

  • Cell number expansion is the first priority in tissue engineering, cell therapy and vaccine manufacturing. Although the 2D cell culture is easy to apply for cell expansion, the issues such as time consuming, labor intensive and the unexpectable cell behaviors need to be defined.
  • Therefore, microcarriers which provide a 3D culture platform and greater surface area for cell adhesion and proliferation, are able to reach the ideal cell number for tissue engineering, cell therapy and vaccine manufacturing.

Tantti® Monolith

Tantti® Monolith has a unique global patent, we call it IMP method (In-Mold Polymerization) and we use In-Mold Polymerization to produce a highly connected honeycomb structure for the production of biological products and other related products. The process is expected to greatly break through the bottleneck of purification for macro molecules, especially accelerate the production of various biologic products such as protein, antibody analysis, exosome and viral particle purification, thereby effectively reducing the cost of protein drugs and vaccine production.
