Application Notes Clone TLD-3A12 has been shown to partially block the proliferative response of antigen-specific CD4+ T cells to antigen-presenting cells and relevant antigen. For Flow Cytometry: Use 10 ul of the suggested working dilution to label 10^6 cells in 100 ul. For IHC-P: The staining of endothelium may be enhanced on FFPE tissue by pre-treatment with 0.2M boric acid pH7.0, See Wilson et al. Immunocytochemistry/Immunofluorescence was reported in scientific literature.

    Background

    CD31, also known as platelet endothelial cell adhesion molecule 1 (PECAM1), is a type I integral membrane glycoprotein and a member of the immunoglobulin superfamily of cell surface receptors. It is constitutively expressed on the surface of endothelial cells and is specifically concentrated at the junction between them. Therefore, CD31 antibodies are commonly used as endothelial markers to measure angiogenesis in association with tumor recurrence. It is also weakly expressed on many peripheral lymphoid cells and platelets. Other studies have also indicated that CD31 and CD34 can be used as markers for myeloid progenitor cells and recognize different subsets of myeloid leukemia infiltrates (granular sarcomas). It is also weakly expressed on many peripheral lymphoid cells and platelets.

    1. I potentially am interested in CD31/PECAM1 Antibody (TLD-3A12). I am woundering if this AB is suitable to use for immunohistochemistry with PFA-fixed section. Is it suitable?
      • This Ab is suitable for IHC with PFA.
    2. What is the size of the target protein CD31/PECAM for this antibody?
      • In western blot, we see bands between 115-160kDa for this particular target. This weight varies from the theoretical molecular weight of 76kDa due to glycosylation of the protein.
    3. What is the concentration NB100-64796?
      • The volume of the antibody provided is 250ul. The concentration of the antibody is 1.0mg/ml.
    4. Can you give me more information about this antibody?
      • Our CD31/PECAM1 mouse monoclonal antibody with catalogue number NB100-64796 was raised against activated Lewis rat derived microglial cells. Since we do not routinely epitope map our antibodies however, I am unable to tell you exactly where on the protein it binds.
    5. Would this antibody apply to tissue that fixed after the formalin? 
      • Formalin fixation should be acceptable for use with this antibody. We incubate freshly dissected tissue in 10% formalin for 4-24 hours, although the incubation time is dependent on tissue type and size. We do not recommend fixation for longer than 24 hours since this can cause epitope masking.
