Karyotype hypertriploid; modal number = 72 to 74. The rate of higher ploidies was 4.2%. Most chromosomes were morphologically normal. Three marker chromosomes were common to all cells: del(1) (q11), der (9)t(7;?;9) (q11?;?;p24), and der (10)t(4;10) (q21;q15)., The der (9) was paired in nearly 50% of the cells. Usually one, but occasionally three double minutes (DM) were seen in a few cells. Five copies of normal N5, N7 and N20 were seen in most cells., The X and Y were paired. The presence of Y chromosomes was confirmed in the QM stained preparation.
Derivation The SW 1088 cell line was initiated by A. Leibovitz at the Scott and White Clinic, Temple, Texas in 1975 from an astrocytoma taken from a 72 year old male Caucasian. The cell line was received at the ATCC in January, 1982 at passage 23.

Complete Growth Medium The base medium for this cell line is ATCC-formulated Leibovitz's L-15 Medium, Catalog No. 30-2008. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.

(Note: The L-15 medium formulation was devised for use in a free gas exchange with atmospheric air. A CO2 and air mixture is detrimental to cells when using this medium for cultivation)

Subculturing

Remove medium, rinse with fresh 0.25% trypsin solution, remove trypsin and let the culture sit at room temperature (or at 37°C) until the cells detach (about 10 minutes). Add fresh medium, aspirate and dispense into new flasks. Subculture every 6 to 8 days. Subcultivation Ratio: A subcultivation ratio of 1:3 to 1:6 is recommendedMedium Renewal: 2 to 3 times per week
