Derivation This is a macrophage-like cell line derived by fusion of normal human peripheral blood leukocytes with the mouse macrophage cell line, RAW 264.
Comments This is a macrophage-like cell line derived by fusion of normal human peripheral blood leukocytes with the mouse macrophage cell line, RAW 264. The cells exhibit chemotaxis to N-formyl-methionine-leucine-phenylalanine (fMet-Leu-Phe, FMLP). WBC264-9 cells express human HPRT.
Complete Growth Medium The base medium for this cell line is ATCC-formulated Eagle's Minimum Essential Medium, Catalog No. 30-2003. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
Subculturing Protocol: Subcultures are prepared by scraping. DO NOT USE TRYPSIN! Remove old medium, add fresh, dislodge cells and dispense into new flasks. Subcultivation Ratio: A subcultivation ratio of 1:2 to 1:6 is recommended Medium Renewal: Every 2 to 3 days
Cryopreservation Freeze medium: Complete growth medium, 95%; DMSO, 5% Storage temperature: liquid nitrogen vapor temperature
