The cells can be sensitized to specific antigens by incubating them with IgE having the desired antigenic specificity.
The cells so coated will release soluble mediators (such as histamine) in response to exposure to the antigen (allergen).

| Subculturing | Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 to 5 X 105 viable cells/ml. Maintain cell density between 2 X 105 and 2 X 106 viable cells/mL.
